The molecular structure and properties of 7-azaindole in its first four singlet states were studied with a view to improving current understanding of the photophysical behavior of its C(2h) dimer. This dimer, which exhibits a double proton transfer via its two hydrogen bonds upon electronic excitation, has for 35 years been used as a model for the photophysical behavior of DNA base pairs. Electronic excitation of 7-azaindole simultaneously increases its acidity and basicity; these changes facilitate a concerted mechanism for the double proton transfer in the dimer. In this work, we found the acidity and basicity changes to occur only in its first pi,pi(*) excited singlet state.  相似文献

LC-MS is a powerful method for the sensitive detection of proteins and peptides in biological fluids. However, the presence of highly abundant proteins often masks those of lower abundance and thus generally prevents their detection and identification in proteomic studies. In human serum the most abundant proteins are albumin and gamma-globulins. We tested several approaches to specifically reduce the level of these proteins based on either specific antibodies, dye ligands (for albumin) and protein A or G (for gamma-globulins). The resulting, depleted serum was analyzed by sodium dodecylsulfate-polyacrylamide gel electrophoresis and LC-MS for the residual presence of these abundant proteins as well as for other serum proteins that should remain after depletion. To test the applicability of this method to real-life samples, depleted serum of a cervical cancer patient was analyzed for the presence of a specific tumor marker protein SCCA1 (squamous cell carcinoma antigen 1; P29508), which is present at ng/ml concentrations. The results demonstrate that SCCA1 can be detected by LC-MS in patient serum following depletion of albumin and gamma-globulins thus opening the possibility of screening patient sera for other, so far unknown, tumor markers.  相似文献

Although frit-fast atom bombardment (frit-FAB) and continuous-flow FAB mass spectrometry have become standard methods for the analysis of peptides and peptide mixtures, these techniques have not been applied previously to the analysis of oligonucleotides. Mobilephase composition, flow rate, and sample size were optimized for the analysis of oligonucleotides by negative ion frit-FAB mass spectrometry (a type of continuous-flow FAB mass spectrometry). With a mobile phase consisting of methanol/water/triethanolamine (80:20:0.5, v/v/w), flow injection frit-FAB analysis of oligonucleotides showed lower limits of detection compared to standard probe FAB mass spectrometry. For example, in order to obtain a signal-to-noise ratio of 3:1, 38 prnol of d(GTIAAC) were required for frit-FAB mass spectrometry and 62 pmol were required for standard probe FAB mass spectrometry. The largest difference between frit-FAB and standard probe FAB was observed for d(pC)5, for which the limit of detection by frit-FAB was approximately 11-fold lower than by standard FAB mass spectrometry. Adjustment of the mobile phase to pH 7 with trifluoroacetic acid increased the limit of detection (reduced sensitivity) a minimum of sixfold. Equimolar mixtures of two or three oligonucleotides produced deprotonated molecules in identical relative abundances whether analyzed by frit-FAB or standard probe FAB mass spectrometry. Finally, frit-FAB liquid chromatography mass spectrometry was demonstrated by separating mixtures of oligonucleotides on a β -cyclodextrin high-performance liquid chromatography column with a mobile phase containing methanol, water, and triethanolamine.  相似文献

Dissolved aluminium in seawater and freshwater is determined by cathodic stripping voltammetry (c.s.v.) preceded by adsorptive collection of complex ions with 1,2-dihydroxyanthraquinone-3-sulphonic acid (DASA) on the hanging mercury drop electrode. Complexation of aluminium by DASA is rapid and no waiting period or heating of the sample is required. Optimal conditions are a DASA concentration of 10?5 M, a solution pH of 7.1–7.3 and an adsorption potential of ?0.9 V; the c.s.v. scan is done in the differential-pulse mode. The limit of detection is 1 nM aluminium for an adsorption time of 45 s. The total time needed, including 50min deaeration and a standard addition, is 10–15 min per sample. No serious interferences were found; u.v. irradiation is recommended for samples containing high levels of organic materials.  相似文献

Theoretical and empirical models can be used to model the migration or separation characteristics in micellar electrokinetic chromatography in order to optimise the resolution. In this paper only empirical models were used, because it is easier and more straightforward to obtain these models. Several empirical approaches for the optimisation of the resolution were compared in order to determine which response should be modelled preferably. The use of models of the effective mobility in combination with average plate numbers proved to be the most suitable approach to optimisation of the resolution, because the relative prediction errors of the models of the effective mobility were a factor of 2-4 smaller than the relative prediction errors of the models of the apparent mobility. Moreover for the least separated peak pair the resolutions based on the models of the apparent and effective mobility showed relative prediction errors that were approximately a factor of 2 smaller than the relative prediction errors of the resolutions based on the models of the resolution and separation factor. The predictions of the separation factor based on the different models generally showed lower prediction errors than the predictions of the corresponding resolutions. Although the relative prediction errors were large, particularly for closely migrating compounds, the empirical approach will probably lead to the optimum separation buffer composition.  相似文献

To investigate the unidirectional rotation of chiral overcrowded biphenanthrylidenes in more detail, the size of the substituent next to the double bond responsible for the unidirectionality of rotation was varied. The thermal and photochemical isomerization of three sterically overcrowded alkenes is described. The behavior of the biphenanthrylidenes with methyl and ethyl substituents is rather similar, and these compounds undergo a unidirectional 360 degrees rotation around the central double bond in a four-step sequence involving two photochemical cis-trans isomerizations and two thermal helix inversions. The only difference between these two true molecular motors was a small entropic effect, which causes the ethyl substituted molecular motor to rotate slightly faster. The behavior of the i-propyl substituted compound differs significantly from that of the other two. Although not all different isomers of the i-propyl substituted molecular motor were detected spectroscopically, experimental data led to the conclusion that this compound can also be considered as a molecular motor and is capable of performing a 360 degrees unidirectional rotation. (1)H NMR and X-ray analysis show a meso-like form as an intermediate in the unidirectional rotation, which proves that the thermal helix inversion is a stepwise process.  相似文献

Compounds of formula [Pb(L2)(NCS)2] (1) and [Pb(L4)(SCN)2] (2) (where L2 is the lariat crown ether N,N'-bis(3-aminobenzyl)-4,13-diaza-18-crown-6 and L4 is the Schiff-base lariat crown ether N,N'-bis(3-(salicylaldimino)benzyl)-4,13-diaza-18-crown-6) were isolated and structurally characterized by X-ray diffraction analyses. The X-ray crystal structures of both compounds show the metal ion coordinated to the six donor atoms of the crown moiety, leaving the corresponding pendant arms uncoordinated. The coordination sphere of lead(II) is completed by two thiocyanate groups that coordinate either through their nitrogen (1) or sulfur (2) atoms. The organic receptor adopts a syn conformation in 1, while in 2 it shows an anti conformation. To rationalize these unexpected different conformations of the L2 and L4 receptors in compounds 1 and 2, as well as the different binding modes found for the thiocyanate ligands, we have carried out theoretical calculations at the DFT (B3LYP) level. These calculations predict the syn conformation being the most stable in both 1 and 2 complexes. So, the anti conformation found for 2 in the solid state is tentatively attributed to the presence of intermolecular pi-pi interactions between phenol rings, for which the dihedral angle between the least-squares planes of both rings amounts to 2.6 degrees and the distance between the center of both rings is 3.766 A. On the other hand, the analysis of the electronic structure has revealed that the Pb-ligand bonds present highly ionic character in this family of compounds. They also suggest a greater transfer of electron density from the NCS- ligands when they coordinate through the less electronegative S atom. The Pb-SCN covalent bond formation mainly occurs due to an effective overlap of the occupied 3p z orbitals of the S atoms and the unoccupied 6p z AO of the Pb atom, while the Pb-NCS bonding interaction is primarily due to the overlap of the 6s and 7s AO of Pb with sp(1.10) hybrids of the N donor atoms. Our electronic structure calculations can rationalize the different coordination of the thiocyanate groups in compounds 1 and 2: the simultaneous formation of two Pb-SCN bonds is more favorable for S-Pb-S angles close to 180 degrees , for which the overlap between the occupied 3p z orbitals of the S atoms and the unoccupied 6 pz AO of the Pb atom is maximized.  相似文献

Crucial steps are a palladium-catalyzed enantioselective imide alkylation, a vinylogous Mannich reaction, and a ring-closing metathesis process. This communication reports the first intercalation into VOPO4·2H2O performed through a solid-state reaction at room temperature. Ethyleneurea, EU (2-imidazolidone) was intercalated into the oxovanadium phosphate by grinding in a mortar stoichiometric amounts of both reagents at room temperature for 30 min. C, H and N elemental analyses of the obtained intercalation compound gave the formula VOPO4·(EU)·H2O. The profile of the absorption bands due to the VOPO4 lattice vibration around 1000 cm−1 was retained after reaction, providing evidence that an intercalation compound and not merely a salt of V-P-O species was produced. For α-VOPO4·2H2O, the interlayer distance is 0.75 nm, whereas for the EU inter-calation compound it is 0.95 nm. The obtained SEM micrographs show that the microstructure of VOPO4·2H2O is changed after reaction, with a total exfoliation of the oxovanadium matrix.  相似文献
